How do you do so "manually"? The only "manual" method is "upload and update" if you don't let Joomla do so itself (and that should be done in a matter of minutes, depending on your server, the update server and internet connection speeds).
I don't know if moving the site between web hosts would influence the update process or cause you to continue to get the update notices - if it's annoying, you can always turn off the email update notification.
